Deniges reagent is prepared by mixing alpha-naphthol in ethanol with potassium hydroxide (KOH) solution. The mixture typically contains 5% alpha-naphthol dissolved in ethanol combined with a strong alkaline solution of KOH. The reagent should be freshly prepared or stored in airtight containers to maintain effectiveness.

Deniges reagent appears as a clear to slightly yellowish alkaline solution with a strong odor of ethanol and high pH due to potassium hydroxide. When added to cultures producing acetoin, the reagent reacts to form a pink to red color, indicating a positive reaction.
🧪 Applications
Deniges reagent is used in microbiology for the detection of acetoin production by bacteria during fermentation. It is part of the modified Voges-Proskauer test and helps differentiate bacterial species capable of producing 2,3-butanediol fermentation products.
