Standard Saline Citrate Solution (2× SSC) is a commonly used buffered saline solution in molecular biology, providing a balanced ionic environment for nucleic acid hybridization and washing steps.
🏭⚗️ Production
This solution is prepared by dissolving 29.2 grams of sodium chloride (NaCl) and 14.7 grams of trisodium citrate dihydrate (C₆H₅Na₃O₇·2H₂O) in one liter of distilled water. The pH is carefully adjusted to 7.0 using hydrochloric acid to ensure optimal buffering capacity. After preparation, the solution is sterilized by filtration and stored at room temperature.
🔬 Properties
The 2× SSC solution is clear and colorless with moderate ionic strength and buffering power. It maintains a neutral pH and supports stable hybridization of nucleic acid strands by maintaining hydrogen bonding during molecular biology assays.
🧪 Applications
Widely applied in Southern blotting, Northern blotting, in situ hybridization, and microarray washing procedures. It is used during hybridization and washing steps to regulate stringency and promote specific binding of nucleic acids.
