Eosin Y-solution 0.5% alcoholic – for microscopy – is a ready to use solution designed for routine staining in the hematoxylin-eosin (H&E) staining process of sample material of human origin. The H&E staining technique is the most commonly used staining method in histology for examining tissue morphology. This product is supplied in a 500 ml bottle, which is sufficient to stain approximately 750 specimens. It is registered as an in vitro diagnostic (IVD) product and carries the CE mark, ensuring compliance with relevant regulatory standards.
🏭⚗️ Synthesis
The hematoxylin-eosin (H&E) staining process relies on a physico-chemical interaction between dyes and cellular components. In the first step, the positively charged hematoxylin dye binds specifically to the negatively charged phosphate groups present in the nucleic acids of the cell nuclei. This results in the nuclei being stained in shades ranging from dark blue to dark violet. Following this, the counterstaining step uses Eosin Y, a negatively charged anionic xanthene dye, which binds to the positively charged proteins in the cytoplasm and extracellular matrix. This interaction causes the cytoplasm and intercellular substances to be stained in varying shades of pink to red. Additionally, erythrocytes absorb the Eosin dye and appear yellow to orange. This differential staining provides clear contrast between nuclear and cytoplasmic components, facilitating detailed microscopic examination.
