🏭⚗️ Production
Fast Green 1% solution is prepared by dissolving 1 gram of Fast Green FCF dye in 100 mL of distilled water or an ethanol–water mixture, depending on the intended application. The solution may be filtered for clarity and stored in an amber bottle to prevent light degradation. It is typically stored at room temperature or refrigerated, based on lab requirements.
🔬 Properties
This reagent appears as a deep green to blue-green liquid. Fast Green is an anionic triphenylmethane dye that binds strongly to basic (positively charged) proteins and cellular structures. It shows maximum absorbance around 625–640 nm depending on the pH and medium. The solution is water-soluble and stable under normal lab conditions.
🧪 Applications
Fast Green 1% solution is widely used in histology as a counterstain in trichrome staining protocols (e.g., Masson’s Trichrome), where it stains collagen or cytoplasm green. It is also used in protein assays, such as the Lowry method, as an alternative or additional stain. In microscopy, it is applied to visualize plant and animal tissues, especially when contrast with other dyes like safranin is needed.
