DPBS is prepared by dissolving precise amounts of sodium chloride (NaCl), potassium chloride (KCl), sodium phosphate dibasic (Na₂HPO₄), potassium phosphate monobasic (KH₂PO₄), calcium chloride (CaCl₂), and magnesium chloride (MgCl₂) in distilled water. The solution is adjusted to a physiological pH of approximately 7.4 and sterilized by filtration or autoclaving.

DPBS is a clear, colorless, isotonic solution with a pH around 7.4. It contains phosphate ions which act as a buffering system to maintain stable pH. The presence of calcium and magnesium ions supports cellular functions such as adhesion and enzyme activity.
