This stain is prepared by dissolving crystal violet in ethanol, then mixing with ammonium oxalate solution. The final mixture usually contains about 2% crystal violet and 0.8% ammonium oxalate in ethanol. The solution is filtered and stored in a dark container to prevent degradation.

Hucker’s Crystal Violet is a purple, water-soluble basic dye that binds strongly to the peptidoglycan layer of bacterial cell walls, especially in Gram-positive bacteria, allowing differentiation during staining.
