Tris Buffered Saline (TABS) with 1% Gelatin is a blocking and washing buffer used in immunoassays and blotting techniques to minimize non-specific binding by saturating potential interaction sites.

🏭⚗️ Production

This solution is prepared by dissolving Tris base (24.2 g/L for 0.2 M) and sodium chloride (8 g/L for 0.137 M) in distilled water. The pH is adjusted to 7.4 using hydrochloric acid. After buffering, 1% (w/v) gelatin is added gradually while heating the solution to around 37–40°C to fully dissolve the protein. The solution is then cooled and filtered to ensure clarity and sterility before use.

🔬 Properties

The solution is clear to slightly opalescent, with a neutral pH and mild viscosity due to gelatin. Gelatin acts as a protein-based blocker that occupies non-specific binding sites on membranes or wells, while Tris and NaCl maintain pH and ionic strength suitable for biological systems.

🧪 Applications

TABS with 1% gelatin is primarily used as a blocking buffer in ELISA, Western blotting, and immunohistochemistry to reduce background signal. It prevents non-specific interactions of antibodies or probes by coating unoccupied sites on plastic or membrane surfaces.

⚠️ Safety

This solution is generally safe under normal laboratory conditions. Use gloves and lab coat during handling to avoid contamination. Prolonged storage may lead to microbial growth; store refrigerated and add preservatives if needed. Dispose of according to standard biological waste procedures.
