This buffer is prepared by dissolving Tris base and EDTA (ethylenediaminetetraacetic acid) in distilled water to achieve the desired molar concentrations, typically 10 mM Tris and 1 mM EDTA. The pH is adjusted to 7.4 using hydrochloric acid or sodium hydroxide. Triton X-100 is then added at a concentration of 0.1% to 1% (v/v), and the solution is mixed thoroughly until homogeneous. The buffer is sterilized by filtration and stored at room temperature or refrigerated depending on use.

The TET buffer is clear and colorless, maintaining a stable pH and providing chelation of divalent metal ions through EDTA. Triton X-100 acts as a mild non-ionic detergent, disrupting lipid membranes and aiding in cell lysis without denaturing proteins.
