TAE buffer is typically prepared by dissolving Tris base, acetic acid, and EDTA (Ethylenediaminetetraacetic acid) in water. The 10X concentrate is made by combining these components at ten times the final desired concentration. Before use in electrophoresis, it is diluted with distilled water to the standard 1X concentration.

🔬 Properties
TAE Electrophoresis Buffer has the chemical formula C₄H₁₁NO₃·C₂H₄O₂ (for the Tris and acetic acid components) and is available as a colorless solution with a slightly acidic pH. The 10X stock solution has a pH of about 8.0, and when diluted to 1X, it maintains a pH of approximately 8.3-8.5. The presence of EDTA helps in chelating divalent cations that could interfere with the stability of DNA or RNA. It is especially suitable for the electrophoresis of large DNA fragments.
